چکیده

The water-soluble glucocorticoid, prednisolone ~l-phosphatc, has been shown to inhibit mitotic activity of normal human leukocytes in short-term culture, with a roughly linear relationship between the log dose of steroid and the percentage decrease in mitosis. Over a concentration range of .002 ug/ml to 10 pg/ml, the mitotic index decreased to 25 per cent of control values. Further studies indicate that most of this inhibitory effect of the steroid operates during the first ~4 hours in tissue culture, although the effect is not demonstrable until the 2d and 3d day, when mitosis begins. The findings indicate that prednisolone does not significantly inhibit mitosis directly, in these cultures, but rather inhibits the conversion of partially differentiated circulating leukocytes to a state capable of mitosis. This action occurs at the same time as that of phytohemagglutinin, which initiates mitosis in these cultures by stimulating the conversion process.

In Vitro Inhibition of Human Sperm Creatine Kinase by Nicotine, Cotinine and Cadmium, as a Mechanism in Smoker Men Infertility

Background Nicotine, cotinine and cadmium are harmful components of cigarettes that have an effect on human reproductive function. Although the effects of cigarette smoke on male reproductive function is characterized in several articles its mechanism of action is still unknown. In the present study, we investigate the effect of nicotine, cotinine and cadmium on human sperm creatine kinase acti...

Potent antiarthritic properties of a glucocorticoid derivative, NCX-1015, in an experimental model of arthritis.

Here, we describe the improved antiarthritic properties of a nitric oxide-releasing derivative of prednisolone that includes a sparing of the effects on bone. Glucocorticoids are widely used in the treatment of chronic inflammatory pathologies, but their use is often accompanied by side effects, including osteoporosis. Recently, a new steroid able to release low levels of nitric oxide showed po...

In vitro Effect of Lead, Silver, Tin, Mercury, Indium and Bismuth on Human Sperm Creatine Kinase Activity: a Presumable Mechanism for Men Infertility

Background: The aim of the present study was to investigate the in vitro effects of mercury (Hg+2), lead (Pb+2), silver (Ag+2), tin (Sn+2), bismuth (Bi+3) and indium (In+3) ions on sperm creatine kinase. Methods: creatine kinase was isolated from human sperm homogenates after chromatography on a DEAE cellulose column. Results: At 60 µg ml-1 metal concentration, 70% of the creatine kinase activi...

In Vitro Evaluation of the Anti-bacterial Effect of Human Platelet Concentrate

Background: Recently the role of platelets in the tissue regeneration, wound healing and prevention and control of infections has been reported. We aimed to assess the antimicrobial effect of human platelet concentrate against six bacteria, commonly found in wound and hospital-acquired infections. Methods: In vitro susceptibility to samples of 10 random human platelet concentrates was determin...
